微生理系统研究进展

    组织工程技术在快速发展30年后,由最初的简单组织构建(例如皮肤的构建)逐步发展到复杂脏器以及微生理系统的制备。在2014年,由美国国家卫生研究院(NIH)、美国国防部研究计划推动局(DARPA)和美国食品和药物管理局(FDA)共同参与、资助了微生理系统的开发及应用工作,包括贝勒医学院、哥伦比亚大学、康乃尔大学、杜克大学、约翰霍普金斯大学、麻萨诸塞州总医院和哈佛医学院、麻省理工学院、西北大学、诺华制药、加州大学欧文分校、中佛罗里达大学、宾夕法尼亚大学、匹兹堡大学、德州大学医疗分部、以及范德堡大学等20多个研究机构参与了这一宏大的计划。


    微生理系统(也常被称为器官芯片)指在体外构建类似人体各种组织及器官间联系的系统,每个系统被设计用来模拟人类器官或内脏器官区域的结构和功能,并且通过微流控系统连接。它们可在体外以极大的生理精度来模拟、研究药物与细胞间,细胞与细胞间,器官与器官间的相互作用。
    根据2014年的论文报导,目前正在进行微生理系统的开发与应用研究,比如在体外模型中的骨和软骨,脑,胃肠道,肺,肝,心脏,微血管,生殖道,骨骼肌,皮肤,和器官芯片之互连来进行生理药物动力学、药物发现和筛选,以及调节干细胞增殖与分化等。创建微生理系统的最初目标是为了提升人类相关药物之研发与测试的效率。该技术着重应用于包括环境毒素对人类的影响,化学品及生化武器之鉴定、控制诱导多功能性干细胞的特定分化,以及人体器官间之代谢与信号转导的动态变化。为了推动微生理系统的发展,我们必须整合多个技术领域,包括微流控芯片、干细胞生物学、三维微结构/矩阵、多细胞工程、各种生物检测技术和资料库工具,以及计算单一或是多个器官系统的电脑模型。微生理系统将革命性的改变基础生物学,生理学,药理学,毒理学和药物,以及一个新的领域:定量系统药理学,因为反复性实验、疾病模型、药效学和药物动力学计算模型是这一新领域的核心。微生理系统应该着重在创造出具有生理意义、简易、可重复、以及具有成本效益的工具来造福整个科学界。
    美国食品药品管理局国家毒理学研究中心主任评论到:“利用微生理系统从生理学、毒理学模拟人体反应来看,确实是有可能的;大量的基础生物学和生理学技术的整合利用也将使我们对于人类健康和疾病过程有更多的了解。”
